  Studies on Defence Gene-Myrosinase Gene from Cruciferae Vegetable

  ABSTRACT

  Glucosinolates-myrosinase is an important defence system in brassica of cruciferous vegetables, also referred to as “the mustard oil bomb”。The myrosinase genes in 49 Brassica parachinensis Bailey、Brassica oleracea L. Var. capitata (L.) and Brassica aboglabra Baily were cloned and analyzed in this paper.

  Three methods of RNA extraction were compared and the results showed that the methods of Phenol-Chloroform and Trizol were better than the method of UNIQ-10. The abundance of RNA in cruciferous vegetable was also related to the attacks from different insects.

  Specific primers based on the the conservative domain of myrosinasewere designed to clone myrosinase genes in 49 Brassica parachinensis Bailey, Brassica oleracea L. Var. capitata (L.) and Brassica aboglabra Baily by RT-PCR.

  One nucleotide acid fragment (350bp) was obtained in Brassica parachinensis Bailey and Brassica oleracea L. Var. capitata (L.) respectively, but two fragments (350bp and 466bp) in Brassica aboglabra Baily.

  It was found that that the fragments of 350bp were highly homologous with the cloned myrosinase genes in Genbank. Therefore, these fragments of 350bp were part of myrosinase genes in these plants. The fragment of 466bp in Brassica aboglabra Baily had an additional nucleotide acid between 310 and 426 compared with myrosinase genes in Genbank. Amino acid sequences were deduced from these fragments and analyzed.

  Primers for RACE were designed based on the nucleotide acid sequences of the fragment that has been cloned in Brassica parachinensis Bailey to gain the full gene of myrosinase (full cDNA). The full myrosinase gene is of 1841bp with an open reading frame of 1647bp. The secondary structure of myrosinase mRNA was predicted with 83 loops that affect the stability of mRNA and the least structure energyof -33.61kcal.

  The protein sequence of 548 amino acids was deduced from this gene. This protein was predicted to be a hydrophilic and dissoluble protein according to the hydrophobicity analyse.

  The secondary structure of the protein was predicted to have 149 amino acids (27.2%)as alpha helixes (Hh), 79 amino acids (14.4%) as beta sheets (Ee), 281 amino acids (51.3%) as random coils (Cc), and 39 amino acids (7.1%) with unknown stuctures(?)。

  The myrosinases in Brassica parachinensis Bailey、Brassica oleracea L. var. capitata (L.) and Brassica aboglabra Baily were found to have a very close relationship through the cluster analyses based on the amino acid sequences.

  Information Granulation Method of Temporal Data and Applications

  ABSTRACT

  In 1965, the famous American computer and control specialist Professor Lotfi A.Zadeh proposed the concept of fuzzy sets, creating a new field of mathematics--fuzzy math. In 1979, Professor Zadeh proposed the concept of information granules, studying a group of similar subjects as a whole or a whole as a part.

  Information granulation gradually become a hot research topic in 1990s.Putting the tool of information granulation into the related fields of data mining and time series processing will undoubtedly broaden the scope of research in the field, with important theoretical and application value.

  This paper focuses on data information granulation methods and applications, giving an overview of fuzzy information granulation model algorithm, and then delve into the application of information granulation in the field of classification, time series, linear prediction, nonlinear prediction, showing the results of information granualation in theory and application. The main contents are as follows:

  A. Information granulation method of temporal data.

  This paper describes three kinds of fuzzy information granulation method: timeline-based information granulation model, information granulation model based on the value axis, taking into account the timeline and numerical information granulation model, and gives specific steps of the algorithm of the corresponding model.

  B. Construct classifier by using information granlation method.

  Construct classifiers using information granulation method through data normalization, fuzzy information granulation, particle selection and etc. Experimental results show an excellent performance of the classifier in both speed and effectiveness on a certain level.

  C. Establish linear forecasting model of temporal data by using information granulation based Two-Stage algorithm.

  Information granulation method was combined with the Two-Stage algorithm, which was given by Chiang Kao and Chin-Lu Chyu(National Cheng Kung University), to establish linear time series forecasting model.

  D. Establsih nonlinear forecasting model of temporal data by using information granulation based vector machine (SVM) algorithm.

  The collection of information granulation methods and support vector machine(SVM) to establish nonlinear time series forecasting model give a full importance of information granulation. Using this model the changes in trends of a time sequence can be given.

  E. Build the information granulation integrated platform.

  Information granulation platform graphical user interface (GUI) was built on the platform of MATLAB. The platform consists of three parts: information supporting platform, the basic platform of the granulation, granulation application platform. Information Granulation platform can achieve the relevant the information granulation of data, and then process data mining.
